The second season continues the final years of the Edo period. Gabimaru, who was the strongest shinobi of Iwagakure, has now fallen and has become a death-sentenced prisoner. He is asked to bring the Elixir of Life from the strange island of the Buddhist paradise, Sukhavati, and all his crimes will be forgiven. With a hope to reunite with his wife, Gabimaru starts his journey along with Yamada Asaemon Sagiri, his assigned executor. There, they learn that many other criminals on death row are also looking for the Elixir. This island is full of man-made statues, unknown creatures, and the hermits who rule this island. Gabimaru must survive these uncertainties and return alive.
